Consano Bio’s C-1101 Receives FDA Fast Track Designation for Chronic Sciatica
FDA Designation Supports Development of Potential Treatment for Chronic Lumbosacral Radiculopathy
Consano Bio, Inc., a clinical-stage biotechnology company focused on treatments for painful orthopedic conditions, announced that the U.S. Food and Drug Administration (FDA) has granted Fast Track Designation to C-1101.
C-1101 is the company’s lead investigational platelet-derived biologic being developed for chronic, painful lumbosacral radiculopathy (LSR), commonly known as chronic sciatica.
The FDA designation comes as Consano Bio continues clinical development of C-1101. There are currently no FDA-approved pharmaceutical therapies specifically indicated for the treatment of chronic LSR.
Why Chronic Sciatica Remains a Major Medical Problem
Chronic lumbosacral radiculopathy is more than ordinary back pain.
The condition happens when spinal nerve roots in the lower back become damaged or irritated. This can cause pain that travels from the lower back into the leg, along with numbness and weakness.
For some patients, the condition can continue for years. In more serious cases, ongoing nerve damage can result in permanent disability.
LSR also creates a significant burden for the healthcare system. Consano Bio estimates that low back pain accounts for around $135 billion in healthcare spending in the United States each year.
The condition is also associated with opioid use. Chronic sciatica can contribute to opioid initiation and long-term opioid use, adding another challenge for patients and healthcare providers.
Patients living with LSR can also experience depression, anxiety and higher rates of suicidal thoughts compared with the general population.
No FDA-Approved Drug Specifically Targets Chronic Sciatica
Despite the large number of people affected by chronic low back and nerve pain, there are currently no FDA-approved pharmaceutical treatments specifically indicated for chronic sciatica.
Existing management approaches may focus on controlling pain or reducing symptoms.
Consano Bio is taking a different approach with C-1101.
The company is developing the treatment to address the biological processes associated with nerve injury rather than simply managing the pain caused by the condition.
What Fast Track Designation Means for C-1101
The FDA grants Fast Track Designation to help speed the development and review of medicines intended to treat serious conditions where there is a significant unmet medical need.
For Consano Bio, the designation provides an opportunity for closer interaction with the FDA during the development of C-1101.
The FDA based the designation on the unmet need associated with chronic LSR and the nonclinical data package submitted by the company supporting the potential of C-1101.
Andrew Hall, chief executive officer of Consano Bio, said the designation highlights the need for new treatment options for people living with chronic sciatica.
He added that C-1101 is being developed to address the underlying biology of nerve injury instead of only managing pain.
C-1101 Uses a Platelet-Derived Biologic Approach
C-1101 is designed as a multi-protein biologic derived from platelets.
The treatment contains controlled concentrations of different biological proteins, including cytokines, growth factors and matrix proteins.
These proteins are intended to support the body's natural repair processes at the site of nerve injury.
The idea is to influence inflammation while also supporting cellular repair.
C-1101 is administered through an epidural injection. The treatment is designed to deliver high concentrations of these proteins directly to the area affected by nerve injury.
Phase 1 Study Is Already Underway
Consano Bio is currently evaluating C-1101 in a Phase 1 clinical study.
The study, known as C-1101-101 and registered as NCT07264270, is a multicenter, randomized, controlled and double-blind trial.
The study is being conducted in symptomatic patients with chronic painful LSR.
Participants are being evaluated across three sequential dose-escalation cohorts. Each participant receives a single epidural injection of C-1101.
The study initially began enrolling patients in Australia.
Following FDA clearance of Consano Bio's Investigational New Drug (IND) application in July 2026, the company is also expanding the study into the United States.
What Researchers Are Looking at in the Study
The early-stage clinical study is designed to provide information about C-1101's safety and how patients respond to different doses.
Because the study is being conducted in sequential dose-escalation groups, researchers can evaluate each dose before moving to higher levels.
The data generated from the Phase 1 study will help guide the future development of C-1101.
The FDA Fast Track Designation also gives Consano Bio an opportunity to work closely with regulators as the program progresses.
C-1101 Could Take a Different Approach to Nerve Injury
Many existing treatments for chronic sciatica focus primarily on controlling symptoms.
C-1101 is being developed with a different goal.
The treatment is designed to act at the site of nerve injury by modulating inflammation and supporting cellular repair. By delivering a combination of biological proteins through an epidural injection, Consano Bio aims to influence the local environment around the damaged nerve.
Whether this approach can translate into meaningful benefits for patients will depend on results from ongoing and future clinical studies.
